Implementing amino acids in manganese-catalyzed biologically inspired oxidations: g-lactonization and ligand design

Abstract

Most of the organic molecules used in chemical industry (and their synthetic precursors)

contain oxygen atoms in their structure. One of the most convenient methods for the

preparation of oxygenated compounds is the direct oxidation of hydrocarbon C-H or C=C

bonds. Currently, the methodologies that are often used to carry out these transformations

rely on the use of stoichiometric oxidants with poor atom economy and second or third row

transition metals. A different scenario is found in metalloenzymes, in which oxidations occur

with high levels of efficiency and selectivity under very mild experimental conditions.

Key to the high efficiency of oxygenases is the well-defined first coordination sphere around

the metal, being the ligands that are directly bound to the metal the ones that have more

impact in its reactivity. Because of that, enzymatic active sites have been taken as source of

inspiration for the development of new oxidation catalysts based on iron and manganese

coordination compounds with N-based tetradentate ligands. In the first part of this thesis, the

main characteristics of these complexes are discussed, exploring the rationale behind the

evolution of their structure (Chapter III). These complexes are then applied in the C-H

functionalization of a-amino acids, in which a general methodology for the preparation of

non-natural analogues based on the g-lactonization of amino acids is presented (Chapter IV).

Site-selective oxidation of 1º, 2º and 3º C-H bonds is obtained in high yields and good to

excellent diastereo- and (where applicable) enantioselectivities.

Another important feature in the structure of oxygenases is the second coordination sphere

around the metal. The large network of amino acid and peptide chains that surround the

enzymatic active site is key to obtain high selectivities, as it can be engaged in non-covalent

interactions between them and with the substrate promoting specific selectivities. In the

second part of this thesis, a step further in the design of artificial catalysts is done, and 2nd

coordination sphere is considered. First, a literature review on oxidation catalysts that use

amino acids in combination with metal complexes is performed (Chapter V), evidencing the

small number of examples reported up to now. With all these precedents, the last chapter of

this thesis describes a catalytic system in which the second coordination sphere is modified

using supramolecular chemistry (Chapter VI). A manganese complex is decorated with an

18-benzocrown-6 ether that interacts with protonated amino acids in the remote position.

Supramolecular recognition allows the proper location of the carboxylic acid moiety to access

the first coordination sphere and get involved in the catalytic cycle. This system permits an

enzyme-like activation of H2O2 using a single acidic residue and the tuning of the

enantioselectivity in asymmetric epoxidations by the modification of the 2nd coordination

sphere

La majoria de molècules orgàniques utilitzades a la indústria química (i els seus precursors

sintètics) contenen àtoms d’oxigen enllaçats a cadenes alquíliques a la seva estructura. Un

dels mètodes més convenients per preparar compostos oxigenats és l’oxidació directa

d’enllaços C-H o C=C. Actualment, les metodologies més utilitzades per dur a terme

aquestes transformacions es basen en l’ús d’oxidants estequiomètrics amb pobra economia

atòmica, selectivitat limitada i l’ús de metalls de la primera o segona sèrie de transició. En

canvi, els metal·loenzims duen a terme reaccions d’oxidació sota condicions experimentals

suaus amb una elevada eficiència i selectivitat.

Un factor decisiu per l’elevada eficiència de les oxigenases és la primera esfera de

coordinació del centre metàl·lic, sent els lligands directament units al metall els que tenen

més impacte en la reactivitat. Per aquest motiu, els centres actius dels enzims s’han

considerat com a font d’inspiració pel desenvolupament de nous catalitzadors d’oxidació

basats en compostos de coordinació de ferro o manganès amb lligands tetraazadentats. A

la primera part de la tesi, es discuteixen les principals característiques d’aquests complexos,

explorant la racionalitat que hi ha al darrere de la modificació de la seva estructura (Capítol

III). Aquests complexos s’utilitzen posteriorment en la funcionalització d’enllaços C-H en aaminoàcids,

per la qual es presenta una metodologia general que permet la preparació

d’anàlegs no naturals mitjançant la reacció de g-lactonització a partir de l’oxidació d’un enllaç

C-H (Capítol IV). L’oxidació regioselectiva d’enllaços C-H 1º, 2º i 3º s’ha obtingut amb

rendiments elevats i bones diastereo- i enantioselectivitats.

Una altra característica important a l’estructura de les oxigenases és la segona esfera de

coordinació al voltant del metall. L’extensa xarxa d’aminoàcids i cadenes peptídiques que

envolten el centre actiu són claus per obtenir elevades selectivitats, ja que poden establir

interaccions no covalents entre elles i amb els substrats, afavorint selectivitats específiques.

A la segona part de la tesi, es fa un pas més enllà en el disseny de catalitzadors i es modifica

també la 2a esfera de coordinació. Primer, es fa una revisió bibliogràfica de catalitzadors

d’oxidació que combinen complexes metàl·lics i aminoàcids (Capítol V), evidenciant els

escassos exemples que es poden trobar a la literatura fins a dia d’avui. Amb aquests

precedents, l’últim capítol de la tesi descriu un sistema catalític en el qual la segona esfera

de coordinació s’ha modificat utilitzant química supramolecular (Capítol VI). Un complex de

manganès s’ha decorat amb un èter 18-benzocorona-6 que interacciona amb aminoàcids Nprotonats

a la posició remota. El reconeixement supramolecular permet la localització idònia

de l’àcid carboxílic per accedir a la primera esfera de coordinació i actuar en el cicle catalític,

aconseguint l’activació del H2O2 amb un únic residu acídic i la modificació de

l’enantioselectivitat en epoxidacions asimètriques gràcies a la modificació de la segona

esfera de coordinació
